The PROBE project is structured into interconnected Work Packages (WPs), designed to drive the project forward by leveraging federated data networks, AI/ML expertise, and multidisciplinary collaboration and ensure a sustained impact on the critical challenges in osteoarthritis (OA) research, treatment, and clinical trial design.

WP1

Lead: Project coordination and management

Led by Erasmus MC & Novartis

Responsible for providing scientific and technical guidance, and professional project management to ensure adequate progress and successful completion of the project.

WP2

Guide: Embedding key stakeholder insights throughout PROBE

Led by Erasmus MC & Sanofi

Will ensure patient and healthcare provider (HCP) involvement and development of regulatory, health technology assessment (HTA), ethical, and privacy governance for compliant and easy access to pre-existing personal health data.

WP3

Create: Development of secure federated database network and foundation models

Led by University of Hamburg & Capgemini

Aims to establish a robust data platform for osteoarthritis (OA) research by integrating datasets with deep and shallow phenotypic profiling.

WP4

Analyse: Identification of predictive biomarkers and clusters of OA patients

Led by Helmholtz Munich & Sanofi

To identify, within and across biomarker modalities: a) baseline prognostic variables for OA progression towards clinically relevant endpoints, b) clusters of patients with OA, which describe distinct molecular endotypes or clinical subgroups c) potential surrogate outcomes for clinically relevant endpoints.

WP5

Implement: Novel enhancements, driven by big data, for optimised and validation-ready clinical trial design, health economics, and new risk-assessment tools to transform OA patient care

Led by University of Leeds & Pacira

Will provide analytical and consensus-based, phase-specific, patient-relevant and stakeholder-accepted models to identify OA subgroups for targeted treatment, and novel models for prediction of disease progression towards clinically relevant endpoints.

WP6

Sustain: Project communication and sustainability

Led by SYNAPSE & Novartis

Aims to promote external communication and disseminate information and outcomes generated by the project to the general public and relevant stakeholders.
